Ensemble's business model centered on a dual strategy: developing its own internal pipeline of drug candidates and forming high-value partnerships with major pharmaceutical companies. These collaborations provided non-dilutive funding and validated the Ensemblin platform. Revenue was generated through upfront payments, research funding, and potential future milestone payments and royalties from these partnerships.
Over its 13-year history, Ensemble secured numerous collaborations with industry leaders including Pfizer, Bristol-Myers Squibb, Boehringer Ingelheim, Novartis, and Alexion Pharmaceuticals. These deals focused on discovering and developing macrocycle candidates for a range of therapeutic areas, primarily oncology and inflammation. The company's internal pipeline included programs targeting IDO and USP9x for cancer, as well as a lead program for an IL-17 antagonist in inflammation, which was later licensed to Novartis. Despite raising approximately $43.5 million in equity financing from backers like Flagship Ventures, Arch Venture Partners, and Oxford Bioscience Partners, Ensemble was unable to advance any of its in-house drug candidates into clinical trials. After failing to secure sufficient ongoing financing, the company ceased operations in late 2017.
