About XTuit Pharmaceuticals
XTuit Pharmaceuticals, founded in 2011, was a biopharmaceutical firm that operated from Waltham, Massachusetts, before ceasing operations. The company focused on creating therapeutics for cancer and fibrotic diseases by targeting the disease-promoting microenvironment. This approach aimed to address issues like hypoxia, drug resistance, and decreased immune response. The business model was centered on its proprietary drug development pipeline and an integrated clinical biomarker platform designed to accelerate the clinical development of its therapies.
The company's intellectual property was built upon the foundational science of its distinguished co-founders: Dr. Rakesh K. Jain of Harvard Medical School and Massachusetts General Hospital, Dr. Robert Langer of MIT, and Dr. Ronald Evans of the Salk Institute. Dr. Jain's research concentrated on the tumor microenvironment and its role in therapy resistance. Dr. Evans' work focused on the regulatory switches that activate stromal cells, which drive fibrosis and tumor growth, while Dr. Langer is a renowned engineer and serial entrepreneur in the biotechnology space. Their collective expertise created the scientific basis for XTuit's core strategy.
XTuit's main product candidates were microenvironment-activated drugs. These compounds were designed to inhibit the synthesis of the extracellular matrix and silence activated stromal cells, such as cancer-associated fibroblasts. By doing so, the drugs could alleviate hypoxia (low oxygen levels in tissues) and reduce solid stress within tumors, thereby enabling better uptake of other therapeutic agents. The company was developing treatments intended to work in conjunction with standard-of-care drugs, including immune checkpoint inhibitors and chemotherapy, to improve outcomes in oncology. Another primary focus was on developing agents to reverse fibrosis in conditions like liver cirrhosis and nonalcoholic steatohepatitis (NASH). The company had not yet reached clinical trials before it became inactive.
After operating in stealth mode for several years, XTuit announced a $22 million Series A financing in June 2015, led by New Enterprise Associates and supported by founding investor Polaris Partners, among others. This funding was intended to advance its lead products into clinical trials. In March 2017, the company appointed Dr. Deborah Dunsire, an experienced leader from major pharmaceutical firms, as its CEO. However, by mid-2018, there were indications the company may have closed, as its website was down and Dr. Dunsire had moved to a new CEO role at H. Lundbeck.
