About DTx Pharma

DTx Pharma, established in 2017 and headquartered in San Diego, operated as a preclinical-stage biotechnology company until its acquisition by Novartis in July 2023. The company was founded by Arthur "Artie" Suckow, John Grundy, and Adam Pavlicek. Dr. Suckow, who also served as CEO, brought a wealth of experience from previous roles at major pharmaceutical companies like Janssen (J&J) and AstraZeneca, with a background in developing various therapeutic modalities. His graduate work at UC San Diego, where he founded a cell therapy company, and his subsequent industry experience informed the central idea behind DTx Pharma: solving the delivery challenges for RNA-based medicines.

The company's core business centered on its proprietary Fatty Acid Ligand Conjugated OligoNucleotide (FALCON™) platform. This technology was developed to overcome a significant hurdle in RNA therapeutics: delivering them to tissues beyond the liver. The FALCON platform attaches fatty acids to small interfering RNA (siRNA), which then bind to albumin in the bloodstream. This process enhances the therapy's biodistribution and cellular uptake, allowing it to reach and act on target genes in the peripheral and central nervous systems, as well as muscle and heart tissues. The business model focused on developing a pipeline of these advanced therapeutics to address the genetic drivers of various diseases, with a primary focus on neuromuscular and central nervous system disorders.

DTx Pharma's lead program, DTx-1252, is a potential first-in-class treatment for Charcot-Marie-Tooth Disease Type 1A (CMT1A), a debilitating inherited neuromuscular disorder with no approved treatment targeting its genetic cause. DTx-1252 works by repressing the PMP22 protein, which is overexpressed in CMT1A patients, and has demonstrated the ability to reverse the disease's effects in preclinical models. This lead asset, which received Orphan Drug Designation from the FDA, and the underlying FALCON platform were key drivers of the acquisition by Novartis. The acquisition was structured with an upfront payment of $500 million and potential milestone payments of an additional $500 million. Prior to the acquisition, DTx Pharma had successfully raised over $115 million from prominent healthcare investors including RA Capital Management, Eli Lilly and Company, and Access Biotechnology.
