About True North Therapeutics

True North Therapeutics was a clinical-stage biotechnology firm established in 2013 as a spin-out from iPierian, Inc. The company was co-founded by CEO Nancy Stagliano, a serial biotech entrepreneur with a Ph.D. in neuroscience. Stagliano previously led iPierian, which focused on neurodegenerative diseases and was acquired by Bristol-Myers Squibb for up to $725 million. Her experience includes co-founding CytomX Therapeutics and a tenure at Millennium Pharmaceuticals, reflecting a career centered on translating scientific discoveries into therapeutic products.

The company focused on developing therapies for rare, complement-mediated diseases in the hematologic, renal, and neurological spaces. True North's business model revolved around the discovery, development, and commercialization of first-in-class product candidates that selectively inhibit the Complement system. Its primary market consisted of patients with rare diseases that had high unmet clinical needs and few, if any, available treatment options.

True North's lead product was TNT009, a monoclonal antibody designed to selectively inhibit the Classical Complement pathway by targeting the C1s protein. This mechanism of action prevents downstream disease processes like phagocytosis, inflammation, and cell lysis, while preserving the essential immune surveillance functions of other complement pathways. The primary indication for TNT009 was Cold Agglutinin Disease (CAD), a rare autoimmune hemolytic anemia where the immune system destroys red blood cells, leading to severe anemia and fatigue. There were no approved treatments for CAD at the time. Clinical trial data for TNT009 was promising, with a Phase 1b study showing that four out of five patients responded within the first day of administration. The drug received Orphan Drug Designation from both the U.S. FDA and the European Medicines Agency (EMA), as well as Breakthrough Therapy Designation from the FDA.

In May 2017, True North Therapeutics was acquired by Bioverativ, a Biogen spin-off focused on rare blood disorders. The deal was valued at $400 million upfront, with an additional $425 million in potential milestone payments. This acquisition provided Bioverativ with a promising clinical-stage asset and strengthened its pipeline beyond hemophilia. Subsequently, in 2018, Sanofi acquired Bioverativ for $11.6 billion, further integrating True North's developments into a global biopharmaceutical company's portfolio.
