Therachon

Therachon, established in 2017 as a spin-off from BioMarin Pharmaceutical Inc., was a clinical-stage biotechnology firm with a focused mission on developing treatments for rare genetic conditions. The company was founded by a group of former BioMarin employees, including its CEO Luca Santarelli, leveraging their extensive experience in the rare disease sector to address achondroplasia, the most common form of dwarfism. Santarelli's background, which includes leading Roche's neuroscience and rare disease units, was instrumental in shaping Therachon's strategic direction and attracting significant investment.

The firm's primary business was centered on the discovery and clinical development of therapeutics for severe and rare diseases. Therachon operated within the orphan drug market, a segment of the pharmaceutical industry dedicated to conditions affecting a small percentage of the population. Its business model revolved around advancing its lead drug candidate through the expensive and complex phases of clinical trials, with the ultimate goal of commercialization or, as is common in the biotech space, being acquired by a larger pharmaceutical company. The company successfully raised $100 million in financing from a syndicate of investors, including OrbiMed, Versant Ventures, and Novo Holdings, to fund its operations and research programs.

Therachon's lead asset was a product candidate named TA-46, later known as Recifercept, a soluble form of the human fibroblast growth factor receptor 3 (FGFR3). This protein was designed to specifically target and inhibit the overactive FGFR3 signaling pathway, which is the underlying cause of the impaired bone growth in individuals with achondroplasia. By acting as a decoy, Recifercept aimed to capture the excess growth factors that would otherwise bind to the mutated receptor, thereby normalizing bone development. This approach represented a targeted therapeutic strategy to address the root cause of the condition. In a significant milestone that validated its clinical strategy and asset value, Pfizer acquired Therachon in 2019 for an upfront payment of $340 million with an additional $470 million in potential milestone-based payments.
