SonoThera is a South San Francisco biotechnology company developing ultrasound-mediated nonviral genetic medicines designed for targeted, redosable delivery of DNA and RNA therapeutics. Co-founded and led by CEO Kenneth Greenberg, Ph.D., its platform pairs RIPPLE ultrasound delivery with PORE payload engineering to enable gene expression, editing and silencing across tissues including skeletal muscle, heart, liver, kidney, adipose and brain, using widely available diagnostic ultrasound systems and commercial contrast agents. Lead programs target Duchenne muscular dystrophy (DMD) and autosomal dominant polycystic kidney disease (ADPKD), with first DMD clinical trials expected in 2027. In June 2026 SonoThera closed an oversubscribed $125 million Series B led by Vida Ventures, with ARK Invest, CureDuchenne Ventures, Leaps by Bayer, Otsuka Pharmaceutical, SymBiosis, UCB Ventures, Vivo Capital and existing investors including ARCH Venture Partners, Illumina Ventures, JJDC, RA Capital and Vertex Ventures HC, to advance lead programs into the clinic and expand the pipeline.
