Semma Therapeutics was established with the specific goal of developing a curative therapy for type 1 diabetes (T1D), a disease affecting millions globally. The company was founded in 2014 by scientist Douglas Melton, alongside Robert Millman and Felicia Pagliuca, and is headquartered in Cambridge, Massachusetts. The motivation for the company's creation is deeply personal for Melton, a professor at Harvard University; his research focus shifted dramatically after his son and later his daughter were diagnosed with T1D. This personal connection is even reflected in the company's name, "Semma," a combination of his children's names, Sam and Emma.

The core of Semma's approach is based on groundbreaking research from Melton's lab at the Harvard Stem Cell Institute, which discovered a method to generate billions of functional, insulin-producing pancreatic beta cells from pluripotent stem cells. This technology was exclusively licensed to Semma to be developed into a cell-based therapy. The business operates in the biotechnology and regenerative medicine market, focusing on creating a treatment that could eliminate the need for daily insulin injections and constant glucose monitoring for individuals with insulin-dependent diabetes. The company's business model revolved around advancing this technology through preclinical and clinical trials to eventually commercialize a therapeutic product. This strategy attracted significant investment, including a $44 million Series A round in 2015 and a $114 million Series B round in 2017, with backers such as MPM Capital, Novartis, and Medtronic.

Semma's therapeutic product is a two-part system. The first component is the large-scale production of stem cell-derived islets (SC-islets) that behave like natural pancreatic islets, capable of producing insulin in response to glucose levels. Preclinical studies in animal models demonstrated that transplanting these cells successfully controlled blood glucose levels. The second, and crucial, component is a proprietary delivery device that encapsulates these cells. This small, implantable device is designed to protect the therapeutic cells from the patient's immune system, which would otherwise attack and destroy them, thereby avoiding the need for long-term, toxic immunosuppressant drugs. In a significant milestone, Vertex Pharmaceuticals acquired Semma Therapeutics in September 2019 for $950 million in cash. Following the acquisition, Semma became a separate operating subsidiary of Vertex, allowing it to leverage Vertex's extensive resources and expertise in bringing transformative medicines to market while continuing its pioneering work under the leadership of its scientific founders.
