About IRX Therapeutics

IRX Therapeutics, Inc., founded in 1994 by John W. Hadden II, was a clinical-stage biopharmaceutical company focused on developing immunotherapies to combat cancer and viral diseases. The company's core mission revolved around reversing the immune suppression often observed in the tumor microenvironment, thereby restoring the body's natural ability to fight the disease. Hadden, a recognized pioneer in immunopharmacology, built the company's product platform on his research aimed at correcting defects in dendritic and T cells. Over its history, the company secured significant funding, raising a total of $190 million across 11 rounds, which included a notable $55.6 million Series A in 2008 and a $33.2 million Series C in 2017. A major milestone was the November 2018 acquisition of its assets by the newly formed Brooklyn ImmunoTherapeutics LLC, which aimed to continue the advancement of cytokine-based immunotherapies.

The company's business was centered on the research and development of novel drugs for physician use in treating tumors and viruses. Its primary asset was IRX-2, a proprietary, cell-derived biologic therapeutic. IRX-2 is a complex mixture of numerous active cytokines, including IL-2, IL-1β, IFNγ, and TNFα, produced by stimulating peripheral blood mononuclear cells from healthy donors. This therapeutic cocktail was designed to restore and activate multiple immune cell types—such as T cells, dendritic cells, and natural killer cells—to enable them to recognize and attack tumors. The business model involved advancing IRX-2 through rigorous clinical trials to gain regulatory approval and collaborating with major research institutions, like Japan's National Cancer Research Center, to explore its potential.

IRX-2 was investigated in numerous clinical trials for a variety of cancers. Its most advanced program studied its efficacy in patients with head and neck squamous cell carcinoma (HNSCC). A Phase 2A trial for HNSCC showed a benefit in overall survival, prompting a larger, global Phase 2B trial known as INSPIRE. The therapy was also evaluated in other solid tumors, including breast cancer, cervical and vulvar intraepithelial neoplasia, and in combination with checkpoint inhibitors like pembrolizumab and nivolumab. The treatment regimen often involved a pre-treatment with low-dose cyclophosphamide, followed by injections of IRX-2. Data from various studies suggested that IRX-2 was generally well-tolerated and could effectively modulate the tumor microenvironment, increase tumor-infiltrating lymphocytes, and up-regulate PD-L1, providing a strong rationale for its use both as a standalone therapy and in combination with other anti-cancer agents.
