This was the first commercial instrument designed to sequence individual DNA molecules directly, a technology Helicos branded as True Single Molecule Sequencing (tSMS). The primary benefit of tSMS was the elimination of the amplification step (like PCR) required by competing technologies. This novel approach reduced biases related to GC content, minimized sample loss, and allowed for the analysis of very small amounts of starting material, making it suitable for sequencing degraded or rare samples. The platform catered to researchers in academic institutions, government labs, and pharmaceutical companies engaged in large-scale genomic studies, such as copy number variation analysis and cancer research. Revenue was generated from the sale of the HeliScope system and proprietary reagents and consumables.
Helicos went public in 2007, raising capital to launch its first commercial product. A significant milestone occurred in 2009 when co-founder Stephen Quake sequenced his own genome using the technology for under $50,000 in reagents. Despite its pioneering technology, the company faced substantial financial challenges and stiff competition from more rapidly advancing sequencing companies like Illumina. These competitive pressures, coupled with high instrument costs and difficulties in increasing read lengths, led to financial distress. Following significant layoffs and a strategic shift towards molecular diagnostics in 2010, the company was delisted from NASDAQ. Unable to secure necessary funding, Helicos BioSciences filed for Chapter 11 bankruptcy in November 2012 and ceased operations.
