About Gate Bioscience

Gate Bioscience is a preclinical biotechnology company founded in 2021, operating from Brisbane, California. The firm is developing a novel class of small molecule therapeutics, trademarked as Molecular Gates, designed to eliminate harmful extracellular proteins at their cellular origin. The company was co-founded by Chief Executive Officer Jordi Mata-Fink, Ph.D., Scientific Co-founder Pat Sharp, Ph.D., and Chief Technology Officer Raman Talwar. Mata-Fink's background includes experience at Flagship Pioneering, where he was involved in launching multiple biotech companies, including Editas Medicine and Rubius Therapeutics. Sharp is credited with being the first to show that small molecules could selectively inhibit protein secretion via the secretory translocon, a key component of Gate's therapeutic approach. Talwar is a serial entrepreneur with a history of building technology companies across various industries.

The company's core technology targets the secretory translocon, the single channel through which the vast majority of the 4,000-plus human extracellular proteins must pass to exit a cell. More than 1,000 of these proteins are implicated in diseases ranging from inflammatory conditions to neurodegeneration and cancers when they are misfolded or produced at incorrect levels. Molecular Gates are designed to bind to this translocon, creating a "gate" that recognizes and blocks a specific disease-causing protein. This blockage prevents the harmful protein from being secreted, redirecting it for natural degradation within the cell, while allowing other beneficial proteins to pass normally. This mechanism offers the advantages of small molecules, such as oral bioavailability and the potential to cross the blood-brain barrier, which is a significant hurdle for biologic therapies like antibodies.

Gate Bioscience operates on a business model centered on drug discovery and development, leveraging its proprietary Molecular Gate Discovery Platform™. This platform integrates a specialized library of molecules with a suite of secretion-focused assays and deep biological expertise to enable rapid and precise drug discovery. The company's financial strategy is underscored by significant venture capital investment. After its founding, Gate Bioscience emerged from stealth mode in November 2023 with a $60 million Series A financing round. The round was co-led by Versant Ventures and a16z Bio + Health, with participation from ARCH Venture Partners and GV. In a major strategic milestone, the company announced a collaboration and license agreement with Eli Lilly and Company in July 2025. This partnership, valued at up to $856 million including an upfront payment, equity investment, and potential milestones, aims to discover and develop Molecular Gate therapeutics for difficult-to-drug proteins, providing Gate with substantial non-dilutive funding and validation of its platform.
