About Endolytix

Endolytix Technology, Inc. is a biopharmaceutical company focused on developing therapeutics for difficult-to-treat antibiotic-resistant infections. Founded on March 3rd, 2020, by Jason Holder, Gary Paul Magnant, and Ron Zwanziger, the company initially incubated at North Shore InnoVentures in Beverly, Massachusetts, before expanding to a custom-built facility. Co-founder Jason Holder, who also serves as Chief Science Officer, is a biotech researcher and life science entrepreneur.

The company is tackling the growing public health challenge of antimicrobial resistance (AMR), particularly in nontuberculous mycobacterial (NTM) infections. The market for NTM disease in the US alone affects an estimated 150,000 to 200,000 patients, with an annual growth rate of 8%. These infections are notoriously difficult to cure due to the bacteria's ability to hide within human macrophage cells, rendering many standard antibiotics ineffective and requiring long, arduous treatment regimens with significant side effects.

Endolytix's core technology circumvents these challenges by using a biological approach rather than traditional small-molecule antibiotics. The company engineers highly specific lytic enzymes, derived from bacteriophages, which are capable of breaking down the complex cell envelope of mycobacteria. To solve the problem of drug delivery, these enzymatic payloads are encapsulated within proprietary liposomes, which are designed to target and penetrate the specific macrophage cells where the bacteria reside. This targeted delivery system enables the enzymes to kill pathogenic mycobacteria, including multidrug-resistant strains, both inside and outside of the host cells. This approach has demonstrated the ability to work in synergy with standard-of-care antibiotics, potentially allowing for lower drug concentrations.

The company's business model is centered on the research, development, and eventual commercialization of these novel biologic drug candidates. Its primary clients will be healthcare providers and patients suffering from chronic respiratory diseases like NTM pulmonary disease (NTM-PD) and tuberculosis. Endolytix has secured significant financing to advance its pipeline, including a $12 million venture round in March 2021 and an additional financing round in October 2024 to progress its lead drug candidate through pre-IND discussions with the FDA. Total funding has reached $21.3 million, with investors including Kineticos Life Sciences Management, Morningside Group, and InnoVentures Lab.
