Vigil Neuroscience is a clinical-stage biotechnology company established in 2020 with a focus on neurodegenerative diseases, specifically by targeting microglia, the brain's resident immune cells. The company was co-founded and incubated by Atlas Venture, launching with an initial $50 million Series A financing round with assets in-licensed from Amgen. The founding President and CEO, Dr. Ivana Magovčević-Liebisch, brought extensive industry experience to the firm, having previously held leadership roles at companies like Dyax and serving on the boards of other biotechnology firms. It was initially developed for Adult-onset Leukoencephalopathy with Axonal Spheroids and Pigmented Glia (ALSP), a rare and fatal genetic neurodegenerative disease. Despite showing promise in early trials, the Phase II IGNITE study for iluzanebart did not meet its endpoints, and the program was discontinued. The second key asset, and the focus of the Sanofi acquisition, is VG-3927, an oral small-molecule TREM2 agonist. This drug is designed for more common neurodegenerative diseases like Alzheimer's, offering the convenience of oral administration and the ability to penetrate the central nervous system effectively. Phase 1 results for VG-3927 in healthy volunteers were positive, demonstrating safety and target engagement, which paved the way for its advancement into Phase 2 trials under Sanofi's direction.
