Triplet Therapeutics

The company focused on developing treatments for repeat expansion disorders (REDs), a category of over 40 genetic diseases such as Huntington's disease, myotonic dystrophy, and various spinocerebellar ataxias. Triplet's scientific premise was based on targeting the DNA Damage Response (DDR) pathway, which human genetic evidence identified as a central driver of the onset and progression of these disorders. The business model centered on creating novel therapies that could halt or slow disease progression by addressing the root genetic cause. Its therapeutic strategy involved developing antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs) to precisely reduce the activity of key components in the DDR pathway. This approach was considered upstream to other methods in development, as it targeted the fundamental driver of the diseases.

Triplet's lead clinical candidate was TTX-3360, an ASO designed to target the MSH3 component of the DDR pathway. The therapy was intended to be delivered via intracerebroventricular injection to reach deep brain structures affected early in Huntington's disease. The company also initiated a natural history study called SHIELD-HD to gather data that would inform its clinical trials. Despite its promising start and scientific approach, Triplet Therapeutics ceased operations around October 2022. The closure was attributed to a combination of factors, including clinical setbacks related to the toxicity of its lead ASO compound in the central nervous system during animal studies, and difficulties in securing further investment. The broader context of clinical trial failures for Huntington's disease by other companies and a general slowdown in biotech financing contributed to the challenging funding environment. Following its closure, the company's assets were acquired through an assignment for the benefit of creditors process.
