AcelRx Pharmaceuticals | Therapies for Acute Pain
About Talphera
Talphera Inc., formerly known as AcelRx Pharmaceuticals, is a specialty pharmaceutical company that has strategically pivoted its focus towards developing and commercializing therapies for use in medically supervised settings. Founded in July 2005 by Dr. Pamela P. Palmer, the company initially concentrated on creating treatments for acute pain. Dr. Palmer, an anesthesiologist with a Ph.D. in neuroscience, dedicated her career to pain research and treatment, holding key positions at the UCSF Pain Center for Advanced Research and Education before co-founding the company. A significant milestone was the FDA approval of its acute pain medication, DSUVIA, a sublingual sufentanil tablet, in November 2018. However, facing commercial challenges, the company divested DSUVIA in March 2023 and rebranded to Talphera in January 2024 to signify a new strategic direction beyond pain management.
The company's business model is now centered on advancing a pipeline of products for hospital and other medically supervised environments. Revenue is not currently being generated from product sales, as the company is in a pre-commercial stage following the divestiture of its previous products. The business strategy relies on securing regulatory approval for its pipeline candidates and forming commercialization partnerships, collaborations, and strategic alliances. The target clients are healthcare providers in settings such as hospitals, surgical centers, and emergency departments.
Talphera's lead product candidate is Niyad™, a lyophilized formulation of nafamostat, designed for use as a regional anticoagulant for the extracorporeal circuit during renal replacement therapy (RRT). This product has received Breakthrough Device Designation status from the FDA and is positioned to be the first approved regional anticoagulant for dialysis circuits, addressing a critical need for patients who cannot tolerate heparin. Nafamostat is a synthetic serine protease inhibitor with a very short half-life, which allows it to act locally in the dialysis circuit without systemic effects. Beyond Niyad, the pipeline includes LTX-608, another nafamostat formulation for potential intravenous use against conditions like acute respiratory distress syndrome (ARDS) and pancreatitis. In partnership with Laboratoire Aguettant, Talphera is also developing two ready-to-use, pre-filled syringes, Fedsyra™ (ephedrine) and PFS-02 (phenylephrine), to streamline the administration of antihypotensive drugs in clinical settings.
