A significant leadership transition occurred in April 2015 when Marco Taglietti, M.D., an infectious disease specialist with extensive experience in drug development at pharmaceutical giants like Schering-Plough and Forest Laboratories, took over as President and CEO after joining the board in 2014. The company went public with an Initial Public Offering (IPO) in 2014 to fund the development of its pipeline.

SCYNEXIS's core business revolves around the research, development, and commercialization of its proprietary class of antifungal agents known as "fungerps". The company's lead product is ibrexafungerp, marketed as BREXAFEMME®, a first-in-class, oral, non-azole treatment. Its mechanism of action, glucan synthase inhibition, is fungicidal, meaning it actively kills fungal cells. The primary market for BREXAFEMME® is patients suffering from fungal infections, initially targeting community-based infections. The U.S. Food and Drug Administration (FDA) first approved BREXAFEMME® in June 2021 for treating vulvovaginal candidiasis (VVC), commonly known as a vaginal yeast infection. This marked the approval of the first new antifungal class in over two decades. A second indication was approved in December 2022 for the reduction of recurrent VVC.

The company's business model has evolved from direct commercialization to a partnership-focused strategy. In March 2023, SCYNEXIS entered into a major exclusive licensing agreement with GSK plc. Under this agreement, GSK acquired the rights to commercialize and further develop ibrexafungerp globally, excluding certain territories. This strategic transaction provided SCYNEXIS with a significant upfront payment of $90 million and the potential for substantial future milestone payments and tiered royalties based on sales performance. This partnership allows SCYNEXIS to leverage GSK's extensive commercial infrastructure while focusing its own resources on advancing its clinical programs. SCYNEXIS continues to execute the Phase 3 clinical trials for ibrexafungerp for more severe, life-threatening invasive fungal infections like invasive candidiasis (IC), which primarily affect hospitalized patients. The company is also developing a second-generation compound, SCY-247, for a range of invasive fungal pathogens.
