The company's strategic objective was to develop disease-modifying therapies that could halt the progression of Alzheimer's by targeting its root causes. Satori's core scientific pursuit involved the creation of small-molecule therapeutics designed to modulate an enzyme known as gamma-secretase. This approach aimed to selectively reduce the production of amyloid beta 42 (AB42) peptides, a primary component of the toxic brain plaques characteristic of Alzheimer's disease, without affecting other necessary cellular functions. The business model was typical for a preclinical biotech firm: secure significant venture capital funding to finance long-term research and development, with the ultimate goal of bringing a drug candidate through clinical trials to market, or achieving a lucrative exit through acquisition by a larger pharmaceutical company.

Satori successfully raised approximately $47.3 million in total funding from prominent life science investors, including InterWest Partners, New Enterprise Associates (NEA), and Prospect Venture Partners. A significant milestone was a $22 million financing round in 2009, followed by an additional $15 million in 2012, intended to advance its lead compounds into clinical development. However, the company's trajectory was abruptly altered in late 2012. Preclinical studies of its lead drug candidate in monkeys revealed unexpected safety issues, specifically the disruption of adrenal function. After exploring various strategic options, the board and investors concluded that the program was no longer viable. Consequently, Satori Pharmaceuticals ceased operations in May 2013, marking another casualty in the challenging quest for an effective Alzheimer's treatment.
