Pleco Therapeutics BV is a clinical-stage specialty biopharmaceutical company, established in the Netherlands in 2018, that is developing therapies to enhance the effectiveness of existing cancer treatments. The company was founded by Ivo Timmermans, who serves as co-CEO. Timmermans has a medical degree and an MBA, with three decades of experience in the pharmaceutical and biotech industries, holding senior roles where he specialized in oncology, immunology, and the strategic development of medicines.

The company's core business revolves around its proprietary Plecoid™ therapies, which are designed to detoxify the cancerous micro-environment. This approach is based on the principle that an accumulation of toxic metals within cancer cells can lead to resistance against chemotherapy. Pleco's products are metal-chelating agents that selectively bind to and remove these toxic heavy metals from the tumor microenvironment, which in turn is expected to improve the function of tumor-suppressing proteins and increase the cancer cells' sensitivity to chemotherapy. The business model focuses on developing and commercializing this pipeline of therapies, initially targeting orphan diseases in oncology. The company collaborates with partners for development and commercialization, such as its co-development of PTX-252 with the Belgian specialty pharma company Hyloris Pharmaceuticals.

Pleco's product pipeline includes several candidates for difficult-to-treat cancers. Its lead programs target Acute Myeloid Leukaemia (AML) and Small-Cell Lung Cancer (SCLC). One of its key candidates, PTX-252, is an intravenous formulation for AML that has received Orphan Drug Designation from the U.S. Food and Drug Administration (FDA). Following a positive Pre-Investigational New Drug (Pre-IND) meeting with the FDA in March 2025, the company has a clearer regulatory path to advance PTX-252 into clinical trials. The company has secured significant funding to advance its pipeline, including a €17.3 million Series A financing round in 2022, which comprised equity from investors like Oost NL, government innovation grants, and project financing from Hyloris Pharmaceuticals. This funding is intended to complete the development of its lead products and prepare for regulatory submissions in the U.S. and Europe. To support its commercialization strategy, Pleco established a U.S. subsidiary, Pleco Therapeutics USA, Inc., in New Jersey.
