Development of treatments for severe neurological diseases
About Pharnext
Pharnext SA was a clinical-stage biopharmaceutical company focused on developing treatments for neurodegenerative diseases with no existing curative options. It was founded in 2007 by Professor Daniel Cohen, a renowned French geneticist recognized as a pioneer in modern genomics and for his work on mapping the human genome. Cohen's background in genomics and his belief that combining existing, approved drugs in low doses could effectively treat complex diseases formed the foundation of the company's research and development approach.

The company's core business revolved around its proprietary R&D platform, Pleotherapy™. This platform utilized big data, genomic analysis, and artificial intelligence to identify and develop synergistic drug combinations, referred to as Pleodrugs™. The business model aimed to reduce the typically long and costly drug development process by repurposing already approved molecules, thus leveraging their established safety profiles. Pharnext's revenue was primarily expected from the commercialization of its drug candidates and potential licensing agreements, though it was largely supported by financing rounds, government subsidies, and R&D tax credits throughout its operational years. The company targeted the global pharmaceutical market for neurodegenerative diseases.
Pharnext's lead product candidate was PXT3003, an oral fixed-dose combination of baclofen, naltrexone, and sorbitol, for the treatment of Charcot-Marie-Tooth disease type 1A (CMT1A), a rare, debilitating inherited neuropathy. PXT3003 had received orphan drug status in both Europe and the United States. However, despite some initially promising results, the pivotal Phase III PREMIER trial failed to meet its primary endpoint in late 2023, largely due to an unexpected improvement observed in the placebo group. A second candidate, PXT864, had shown encouraging Phase II results for Alzheimer's disease but its development was not advanced.
Following the clinical trial setback and subsequent failure to secure necessary financing, Pharnext faced severe financial distress. The company initiated a drastic cost-cutting plan in early 2024 and its shares, once listed on the Euronext Growth stock exchange in Paris, were suspended. In mid-2024, Pharnext announced it was unable to continue its operations and requested to enter judicial liquidation, which was officially ordered by the Paris Commercial Court in August 2024.
