KAI Pharmaceuticals
Discovers and develops novel therapeutics for cardiovascular diseases, kidney diseases and pain
About KAI Pharmaceuticals
KAI Pharmaceuticals, Inc. was a clinical-stage biopharmaceutical company established in 2002 by founder Daria Mochly-Rosen. Headquartered in South San Francisco, the company focused on developing treatments for cardiovascular and kidney diseases. Its operations centered on discovering and advancing novel drug candidates to address significant unmet medical needs.
The company's primary business was the research and development of pharmaceutical agents. KAI's lead product candidate was KAI-4169, a peptide agonist of the calcium-sensing receptor designed to treat secondary hyperparathyroidism (SHPT) in patients with chronic kidney disease (CKD) undergoing dialysis. This condition is a serious and common complication for these patients. The business model involved significant investment in clinical trials to prove the safety and efficacy of its therapeutic candidates, with the ultimate goal of obtaining regulatory approval and commercializing the product. Revenue generation was projected through drug sales and potential licensing agreements. In September 2011, KAI entered into a partnership with Ono Pharmaceutical Co., Ltd. for the development and commercialization of KAI-4169 in Japan.
KAI-4169, which later became known as Etelcalcetide, was administered intravenously during dialysis sessions. Its key benefit was the demonstrated ability to cause sustained reductions in parathyroid hormone (PTH), phosphorus, and calcium levels, which are critical markers for SHPT. The company successfully navigated the drug through multiple clinical phases, including a Phase 2 study that showed promising results and good tolerability among patients. This progress culminated in a major milestone on April 10, 2012, when Amgen announced its acquisition of KAI Pharmaceuticals for $315 million in cash, gaining worldwide rights to KAI-4169, excluding Japan. Following the acquisition, KAI became a wholly owned subsidiary of Amgen, which continued the development of the drug. In February 2017, the FDA approved Etelcalcetide under the trade name Parsabiv for the treatment of SHPT.
