Heartseed Inc. is a clinical-stage biotechnology company headquartered in Tokyo, Japan, that specializes in developing regenerative therapies for heart disease. The company was founded in 2015 by Dr. Keiichi Fukuda, a renowned professor and global leader in cardiac regeneration research from Keio University School of Medicine. Dr. Fukuda's journey began in the 1990s, and in 1999, he achieved a world-first by successfully creating cardiomyocytes from bone marrow stem cells. Heartseed's core business revolves around its lead product, HS-001, which consists of allogeneic iPSC (induced pluripotent stem cell)-derived, highly purified ventricular cardiomyocyte spheroids. These tiny clusters of heart muscle cells are designed to be transplanted directly into a patient's damaged heart tissue. The proposed mechanism of action, termed "remuscularization," involves the transplanted cells engrafting and electrically coupling with the host's myocardium to improve the heart's contractile function. This approach aims to provide a fundamental cure for severe heart failure, a condition where treatment options are often limited to heart transplants or ventricular assist devices.

The company targets patients with severe heart failure, including those with conditions like Ischemic Heart Disease, Dilated Cardiomyopathy (DCM), and Old Myocardial Infarction (OMI). Its business model is centered on the research, development, and commercialization of these cell therapies. A significant milestone was achieved in June 2021 when Heartseed entered into a worldwide collaboration and license agreement with Novo Nordisk. Under this agreement, Novo Nordisk acquired exclusive rights to develop, manufacture, and commercialize HS-001 globally, except in Japan, where Heartseed retains development rights and shares commercialization costs and profits with Novo Nordisk. This partnership provided Heartseed with substantial funding, including an upfront payment of $55 million, with total potential payments reaching up to $598 million, plus royalties.

Heartseed has been advancing HS-001 through clinical trials. The LAPiS Study, a Phase I/II trial in Japan, has completed patient enrollment to evaluate the safety and efficacy of the therapy when administered during open-heart surgery. The company is also developing less invasive methods, such as transendocardial injection using a catheter system (HS-005), in partnership with companies like Japan Lifeline. Having raised over $76.7 million through multiple funding rounds and successfully listing on the Tokyo Stock Exchange Growth Market in July 2024, Heartseed is positioned to continue its pioneering work in cardiac regenerative medicine.
