ARCA's lead product candidate was Gencaro (bucindolol hydrochloride), a genetically-targeted beta-blocker being developed for the prevention of atrial fibrillation in patients with heart failure. Gencaro had a prolonged and complex development history, having initially been developed for hypertension and heart failure in the early 1990s by Bristol Myers Squibb. After failing a large NIH-sponsored trial, Dr. Bristow and ARCA resurrected the drug by identifying a specific genetic variation that appeared to predict a patient's positive response, aiming for it to be the first genetically targeted cardiovascular therapy. The company also had another product candidate, rNAPc2 (AB201), which was being evaluated as a potential treatment for COVID-19 and other diseases caused by RNA viruses.
In a significant strategic shift, ARCA biopharma announced a merger with Oruka Therapeutics, Inc. in April 2024. The merger was completed on August 29, 2024, after which Oruka became a wholly-owned subsidiary of ARCA. Following the merger, the combined entity began operating under the name Oruka Therapeutics, Inc., and its stock commenced trading on Nasdaq under the new ticker symbol "ORKA" on September 3, 2024. This transaction marked the end of ARCA biopharma as an independent entity and shifted the new company's focus to Oruka's pipeline, which is centered on developing novel biologics for chronic skin diseases like plaque psoriasis. As part of the agreement, pre-merger ARCA stockholders received a special cash dividend and retained a small percentage of ownership in the new combined company.
