About AlloCure
AlloCure, Inc. operated as a clinical-stage biotechnology company with a focus on developing treatments for kidney disease, particularly acute kidney injury (AKI). Founded in 2008 and based in Burlington, Massachusetts, the company has since been marked as deadpooled. AlloCure secured significant venture financing, raising a total of $47 million over five rounds. This included a $14.5 million Series A in July 2008, led by SV Life Sciences and Novo A/S, and a $25 million Series B in 2012, which added Lundbeckfond Ventures to the syndicate.
The company's business model was centered on the research and development of a proprietary cell therapy, with the goal of commercializing a treatment for a significant unmet medical need. The primary therapeutic candidate was AC607, an off-the-shelf, allogeneic mesenchymal stem cell (MSC) therapy. This therapy was developed to be administered to any patient without the need for tissue or blood typing. The cells were sourced from the bone marrow of healthy adult donors and then expanded through a manufacturing process. The business strategy involved progressing AC607 through rigorous clinical trials to gain regulatory approval, which was supported by the FDA granting it Fast Track designation.
AlloCure's product, AC607, was designed to address AKI by leveraging the regenerative capabilities of MSCs. The proposed mechanism of action involved the cells migrating to the site of injury in the kidney, where they would secrete beneficial paracrine factors. This process was intended to promote anti-inflammatory responses and instruct the damaged tissue to repair itself, rather than developing fibrosis, without the stem cells differentiating and repopulating the kidney. The therapy progressed to a multi-center, randomized, double-blind, placebo-controlled Phase 2 clinical trial involving patients who developed AKI after cardiac surgery. However, the trial did not show a significant difference in the time to recovery of kidney function between the group receiving AC607 and the placebo group.
