AAVLife, initially registered in Paris as AAVLife SAS and later known as Annapurna Therapeutics, was a biotechnology firm established in 2014. The company was co-founded by a team of experienced researchers and executives, including CEO Amber Salzman, a former R&D executive at GlaxoSmithKline with a background in managing large-scale clinical trials. Other founders included prominent figures in gene therapy such as Patrick Aubourg, M.D., Ph.D., Pierre Bougnères, M.D., Ph.D., and Ronald Crystal, M.D., who served as a scientific adviser. Dr. Salzman's journey was deeply personal, driven by her experience with her son's rare disease, which fueled her urgency to translate scientific research into clinical applications.

The firm concentrated on developing adeno-associated virus (AAV) mediated gene therapies for rare diseases with significant unmet medical needs. Its primary focus was on creating a treatment for the cardiac dysfunction associated with Friedreich's ataxia, a progressive genetic disorder that affects the neurologic and cardiac systems, often leading to death in early adulthood. The company's core technology, based on research from the French National Institute of Health and Medical Research (Inserm), involved using a harmless AAV vector to deliver a normal frataxin gene into cardiac tissue to restore heart function. This approach showed success in preclinical mouse models, reversing pathological heart enlargement. In addition to Friedreich's ataxia, the company's pipeline expanded to include programs for Alpha1-antitrypsin (A1AT) deficiency, hereditary angioedema, and severe allergies through a collaboration with Weill Cornell Medicine.

The business model centered on advancing its gene therapy candidates through preclinical and clinical trials, with the ultimate goal of commercialization. AAVLife secured $12 million in a Series A financing round in April 2014, led by Versant Ventures and Inserm Transfert Initiative, to fund its research and development activities. In a significant milestone, AAVLife was acquired by the U.S.-based gene therapy company Avalanche Biotechnologies in a definitive agreement announced in February 2016. This transaction was structured to combine Avalanche's ophthalmic programs with Annapurna's rare disease pipeline, creating a more robust entity in the gene therapy space. Following the acquisition, the merged company was renamed Adverum Biotechnologies in May 2016.
